Abstract
Daily calorie restriction (CR) and intermittent fasting (IF) enhance longevity and cognition but the effects and mechanisms that differentiate these two paradigms are unknown. This study compared IF (every-other-day feeding) to a matched 10% daily CR and ad libitum conditions in female C57BL6 mice. After 3 months, IF improved long-term memory retention, increased hippocampal neurogenesis (AHN), and upregulated the longevity gene *Klotho* (*Kl*) in the hippocampus. Downregulating *Kl* in human hippocampal progenitor cells decreased neurogenesis, while overexpression increased it. *Kl* knockout mice showed impaired AHN. The findings suggest IF is superior to 10% CR for enhancing memory, and identify *Kl* as a key regulator of IF's effects on cognition through AHN.
