Evaluation of the Impact of Concentration and Extraction Methods on the Targeted Sequencing of Human Viruses from Wastewater

Sequencing human viruses in wastewater faces low-abundance challenges; this study compared four concentration/extraction methods (Innovaprep, Nanotrap, Promega, Solids) and found method-dependent virus profiles. Innovaprep ultrafiltration yielded the highest sequencing sensitivity and assembled near-complete genomes, while Promega and Nanotrap were more sensitive for SARS-CoV-2 by dPCR.
