Abstract
Library adaptors are short oligonucleotides attached to RNA and DNA samples for next-generation sequencing (NGS). This paper introduces Control Library Adaptors (CAPTORS), which measure NGS accuracy and reliability. Integrated into library preparation, CAPTORS' encoded information is retrieved during sequencing. The authors demonstrate CAPTORS' use in measuring nanopore sequencing accuracy, evaluating metagenomic and RNA sequencing performance, and improving sample normalization. Customization for clinical diagnoses is also shown, correcting systematic sequencing errors and improving breast cancer *BRCA1/2* variant diagnosis. CAPTORS offer a simple and effective method to enhance NGS accuracy and reliability.
