We developed an all-optical approach to probe synaptic plasticity at identified cerebellar synapses in awake, behaving mice, using JEDI-2Psub to record Purkinje cell dendritic activity while selectively activating granule cell and climbing fiber inputs, revealing how pairing granule cell activation with sensory-evoked climbing fiber inputs triggers long-term plasticity of inhibitory responses in Purkinje cells.
