Abstract
This study investigated the role of gut microbiota in high-methionine (HM) diet-induced hyperhomocysteinemia (HHcy) and glucose intolerance in mice. Results showed that HM diet induced HHcy and glucose intolerance, while antibiotic treatment reduced plasma Hcy levels and reversed glucose intolerance. Gut microbiota depletion did not affect liver homocysteine metabolism but altered microbiota composition, increasing *Faecalibaculum* and *Dubosiella*, which were positively correlated with plasma Hcy. *Dubosiella newyorkensis* produced Hcy in vitro. The findings suggest that the HM diet-induced HHcy and glucose intolerance are mediated by gut microbiota-produced Hcy.
