While axolotls can fully regenerate amputated limbs, their bone fracture healing mechanism is unclear. This study presents a surgical technique to stabilize axolotl femur osteotomies using a fixator plate, comparing it to non-stabilized osteotomy and limb amputation. Plate-fixated femurs healed more efficiently. Similar to mice, PCNA+ proliferating cells and SOX9-expressing cells were observed in early stages, suggesting endochondral ossification as the healing mechanism. This standardized fixation technique enables controlled axolotl bone healing experiments comparable to mammals.
