This study presents a universal integrated platform for the streamlined and on-demand preparation of mRNA products directly from DNA templates. The platform incorporates PCR amplification of target DNA, in vitro transcription (IVT) into mRNA, magnetic purification, and encapsulation in lipid nanoparticles (LNPs). Using enhanced green fluorescent protein (eGFP) as a model, the platform achieved a production rate of 200–300 µg of eGFP mRNA in 8 h. Furthermore, an mRNA vaccine encoding the receptor-binding domain (RBD) of SARS-CoV-2 spike protein was successfully produced. This platform offers advantages for universal and on-demand mRNA product preparation, facilitating broader access to mRNA technology and enabling rapid development of mRNA-based vaccines and therapies.
