This study developed highly sensitive SARS-CoV-2-specific antibody and neutralization assays. All PCR+ subjects (n=115) showed SARS-CoV-2 Spike or Nucleocapsid protein-specific IgG antibodies, while negative controls did not. Neutralization was observed in COVID-19 and convalescent plasma. Hospitalized patients had significantly higher antibody and neutralization titers than outpatients or convalescent plasma donors. Some COVID-19 patients also had neutralizing antibodies against SARS-CoV. The assays' high specificity and sensitivity could improve understanding of the antibody response in COVID-19 and vaccine effectiveness.
