Abstract
Rapid construction of diverse genetically encoded biosensors remains challenging. This paper presents GECFINDER, a method to rapidly convert enzymes into biosensors. By inserting a circularly permuted fluorescent protein (cpFP) into the hinges of ANL superfamily enzymes, ligand-induced conformational changes are translated into fluorescence signal changes. The authors generated 11 GECFINDERS for various organic and amino acids, demonstrating their efficacy in quantifying target molecules in vitro. GECFINDER simplifies amino acid quantification, showing promise for point-of-care testing and high-throughput screening in industrial applications.
