Innate lymphoid cells (ILCs) are important immune regulators, but their precise tissue localization and microenvironmental interactions remain unclear. This study combines multiplexed immunofluorescence and a computational analysis pipeline to identify CD127+ ILCs *in situ*, characterize them, and define their microenvironments. The researchers identified IRF4 as a marker for tonsillar ILC3s and conserved stromal landmarks for ILC localization, revealing shared niches with plasma cells in the tonsil. This work provides a platform for multiparametric histological analysis of ILCs to advance ILC biology understanding.
