Abstract
Conflicting data exists on the role of branched-chain amino acids (BCAAs) in metabolic health. This study investigated the individual effects of leucine and valine in mice fed high-fat diets. Leucine supplementation showed beneficial effects on adiposity and insulin sensitivity, while valine worsened high-fat diet-induced health impairments, reducing glucose tolerance and insulin sensitivity due to increased 3-hydroxyisobutyrate (3-HIB) levels, which increased basal skeletal muscle glucose uptake, leading to glucotoxicity and impaired myocyte insulin signaling.
