Abstract
This study identifies ApoL6 as a lipid droplet (LD)-associated protein primarily found in adipose tissue, specifically adipocytes. ApoL6 expression is low during fasting but increases upon feeding. ApoL6 knockdown reduces LD size and triglyceride (TAG) content, while overexpression increases LD size and TAG content. The study demonstrates that ApoL6 inhibits lipolysis by directly interacting with Perilipin 1 (Plin1) and preventing its interaction with hormone-sensitive lipase (HSL). ApoL6 ablation reduces white adipose tissue mass and protects against diet-induced obesity, whereas ApoL6 overexpression leads to obesity and insulin resistance, suggesting ApoL6 as a potential therapeutic target for obesity and diabetes.
