Abstract

This research optimized the RL-QN15 peptide to create CYRL-QN15, a cyclic heptapeptide with improved efficacy in treating skin wounds. A Zn²⁺-crosslinked sodium alginate (ZA) hydrogel containing hollow polydopamine (HPDA) nanoparticles loaded with CYRL-QN15 (HPDA/CYRL-QN15/ZA hydrogel) was developed. This hydrogel accelerated skin cell proliferation, migration, and tube formation, regulated cytokine secretion from macrophages, scavenged free radicals, and reduced reactive oxygen species. In diabetic mice, the hydrogel significantly accelerated full-thickness skin wound healing by promoting M2 macrophage polarization, re-epithelialization, granulation tissue formation, collagen deposition, and angiogenesis. The hydrogel also showed efficacy in treating diabetic skin ulcers in patients. The HPDA/CYRL-QN15/ZA hydrogel offers a novel therapeutic strategy for chronic skin wound healing.
