This study synthesized a comprehensive library of 32 PNAG pentasaccharides to identify PNAG sequences with specific free amine patterns as epitopes for vaccines against *Staphylococcus aureus*. Active and passive vaccination using a conjugate of the discovered PNAG epitope with mutant bacteriophage Qβ provided near-complete protection against *S. aureus* infections in mice, including MRSA. This library is a valuable tool for next-generation vaccine design.
