Abstract
This paper presents SCOPE, a single-step CRISPR-based diagnostic platform for rapid and sensitive detection of monkeypox virus (MPXV). SCOPE uses a streamlined viral lysis protocol and a 10-minute single-step recombinase polymerase amplification (RPA)-CRISPR/Cas13 reaction, conducted within a portable device. The platform can detect as few as 0.5 copies/µL of MPXV within 15 minutes and showed 100% concordance with real-time PCR in a clinical sample validation study (n=102).
