This study demonstrates the applicability of single-cell T-cell receptor sequencing (scTCRseq) in dogs with cancer. Researchers profiled PBMCs or lymph node aspirates from healthy dogs, dogs with T-cell lymphoma, and dogs with melanoma using scRNA and dog-specific scTCRseq primers. Healthy dogs showed highly diverse repertoires, lymphomas exhibited clonal repertoires, and melanoma dogs displayed a limited number of abundant clonotypes. Analysis of dominant clonotypes in melanoma PBMCs revealed predominantly activated CD8+ T cells, suggesting potential anti-tumor activity.
