Abstract
This paper introduces Gut Microbiome Wellness Index 2 (GMWI2), an improved version of GMWI, designed as a disease-agnostic health status indicator based on gut microbiome taxonomic profiles. Analysis of 8069 stool shotgun metagenomes from 54 studies showed GMWI2 achieved 80% cross-validation balanced accuracy in distinguishing healthy from non-healthy individuals, exceeding the original GMWI and traditional α-diversity indices. Inter-study and external validation maintained ~75% accuracy. GMWI2, available as an open-source tool, provides a resource for evaluating health using individual gut microbial composition.
