Abstract
This study presents a high-throughput 4D TIMS lipidomics platform for clinical profiling of circulatory lipids. Automated lipid extraction, combined with microflow reversed-phase UHPLC-TIMS-MS and PASEF, enables high-throughput analysis. A stringent 4D annotation strategy and TOF MS quantification using level-3 internal standards allow reproducible lipid profiling in NIST human plasma and serum. The platform is applied to profile intra-individual lipidome phenotypes in various blood matrices, demonstrating its utility for high-throughput clinical applications.
