This research paper details the development and validation of CoV2-ID, a rapid and highly sensitive five-plex RT-qPCR assay for the detection of SARS-CoV-2. The assay targets three viral genes (Nsp10, Nsp12, and N), includes a human internal control, and an artificial RNA control for quality assessment. CoV2-ID demonstrates 100% sensitivity and specificity, with a limit of detection of a single viral RNA copy and a run time of approximately 15 minutes. The assay also incorporates a mutation detection component for the D614G mutation.
