Abstract
Miscanthus, a rhizomatous perennial plant, has great potential for bioenergy production due to high biomass and stress tolerance. This study reports a chromosome-scale assembly of the Miscanthus lutarioriparius genome using Oxford Nanopore sequencing and Hi-C. The 2.07-Gb assembly covers 96.64% of the genome with a contig N50 of 1.71 Mb. Centromere sequences were assembled for all 19 chromosomes, and telomere sequences for chromosome 10. Two centromeric satellite repeat types confirm an allotetraploid origin, and sorghum-aligned synteny reveals tetraploid genome structure and several chromosomal rearrangements. Tandem duplicate genes are functionally enriched for stress responses and cell wall biosynthesis. Gene families related to disease resistance, cell wall biosynthesis, and metal ion transport are greatly expanded and evolved, suggesting genomic bases for the notable traits of M. lutarioriparius.
