This paper introduces the next-generation mesoSPIM, a benchtop light-sheet microscope designed for imaging large cleared tissues. The improved mesoSPIM features a significantly increased field of view, improved resolution, higher throughput, lower cost, and simpler assembly compared to the original version. It achieves high spatial resolution (1.5 µm laterally, 3.3 µm axially) and supports sample sizes ranging from sub-mm to several centimeters. The microscope is versatile and applicable across multiple fields, including neuroscience, developmental biology, pathology, and physics.
