Cancer immunotherapy, while a significant advancement, faces challenges like drug resistance. This review explores how smart drug delivery systems (SDDSs) can overcome this resistance. SDDSs are designed for precise drug delivery, boosting efficacy by increasing drug concentration at target sites and improving immune responses. The authors discuss how SDDSs overcome resistance by combining immunogenic cell death with immunotherapy, reversing immunosuppressive tumor microenvironments, modulating the interferon signaling pathway, and improving cell therapy efficacy. Future perspectives on SDDS development for enhanced immunotherapy are also presented.
