Abstract
Despite ongoing vaccine and therapeutic development, SARS-CoV-2 continues to evolve. Nurse sharks immunized with SARS-CoV-2 antigens produced IgNAR antibodies targeting conserved spike RBD epitopes. These shark antibodies (ShAbs) potently neutralized SARS-CoV-2 variants, including Omicron, and protected mice from SARS-CoV-2 challenge. Structural analysis enabled the design of multi-specific nanobodies with enhanced neutralization and picomolar affinity to sarbecoviruses. These shark nanobodies are potential immunotherapeutics.
