Abstract
This paper proposes a method for real-time NMR of hyperpolarized proteins at residue resolution using dissolution dynamic nuclear polarization (d-DNP). Hyperpolarized buffers selectively boost NMR signals of solvent-exposed protein residues, enabling residue-resolved spectra at a 2 Hz sampling rate. This addresses limitations of NMR experiments performed under non-physiological conditions and the impediment of long signal accumulation to fast real-time monitoring, crucial for studying protein kinetics and pharmacological relevance.
