Abstract
This study investigated the role of lysine crotonylation in liver cancer progression, focusing on the impact of hypoxia. Researchers found that crotonylation was upregulated under hypoxic conditions and promoted liver cancer cell growth. Through quantitative lysine crotonylome analysis, they identified 3,793 crotonylation sites in 1,428 proteins. They demonstrated that lamin A crotonylation at K265/270 maintains its subcellular localization, promoting cell proliferation and preventing senescence. HDAC6 was identified as the lamin A decrotonylase, downregulated under hypoxia, leading to increased lamin A crotonylation. The study highlights the role of lamin A crotonylation in liver cancer progression and expands understanding of non-histone protein crotonylation.
