The threat of coronavirus spillovers necessitates vaccines offering broader sarbecovirus protection. A single antigen, computationally designed using the receptor-binding domain (RBD) of the spike protein, elicited broad humoral responses against SARS-CoV-1, SARS-CoV-2, WIV16, and RaTG13 in mice, rabbits, and guinea pigs. Administered as DNA or MVA, this antigen provided protection against the Delta variant in ACE2-expressing mice primed with AZD1222. mRNA delivery further validated its broad immunogenicity, highlighting its potential to counteract zoonotic betacoronavirus threats.
