Abstract
Cryo-electron tomography (cryo-ET) combined with subtomogram averaging (StA) is a powerful technique for solving macromolecular structures in situ. However, its widespread adoption is hindered by the complexity and time-consuming nature of data processing. TomoBEAR, a modular and configurable workflow engine, is presented to streamline cryo-ET data processing for StA. It integrates commonly used cryo-EM packages with sensible presets, enabling high-throughput processing and minimizing manual intervention. Applications to various datasets demonstrate TomoBEAR's ability to produce high-resolution structures, accelerating the adoption of in situ structural biology.
