Abstract
African swine fever virus (ASFV) is a highly contagious and lethal virus affecting pigs, causing significant economic losses. This paper describes the development of a rapid, sensitive, and portable Cas12a-mediated paper assay for ASFV detection. The assay uses crRNAs targeting highly conserved regions of ASFV genes, demonstrating high specificity and low tolerance for mismatch mutations. A paper-based lateral flow strip, combined with Cas12a detection, enables instrument-free detection with sensitivity comparable to qPCR.
