This study aimed to improve the miRNA isolation process from cultured cells using a commercially available extraction kit. The researchers investigated factors influencing RNA adsorption (ethanol concentration, Ca²⁺ addition) and elution (temperature, pH). Optimal conditions were determined as 65% (v/v) ethanol for adsorption and TE buffer (pH 8.0) at 55°C for elution. The modified method increased endogenous miR-21 recovery approximately sixfold compared to the original kit.
